The EjacuChip

The Shredder is a microfluidic dissection device that uses a fine glass needle to slice through the cuticle and testis of males to release stored spermatids.

The Pheromone Arena is designed to quantify pheromone communication and choice using real time production of pheromones in a controlled environment.

The EjacuChip sequentially loads up to 80 males into individual channels. The chip was designed to induce ejaculations for sperm collection, however, it is also amenable for single worm imaging.

Worm Strains

The Kasimatis lab designation is KRK and allele designation is uva.

NSPF gene family: Deletion and transgene

The NSPF deletion allele (fxDf1) removed nspf-1, nspf-2, and nspf-3 using CRISPR.
To created a tagged trans gene, the promoter sequence for nspf-1 along with a 6xHis-tagged nspf-1 were inserted into the nspf deletion allele background (PX814) using CRISPR.

Inducible male sterility

Auxin-inducible spermatogenesis arrest, resulting in hermaphrodite and/or male self-sterility. Degron tag was inserted into the endogenous spe-44 locus using CRISPR.

Experimental Evolution of Male Reproductive Success

These strains were created in Kasimatis et al. 2022 for isolating evolution of sperm competitive ability. Other strains created in this study are available from the Phillips lab upon request.
